چکیده

The control of gene regulation can be examined by somatic cell hybridization. Hybrids between human cell lines belonging to different hematopoietic lineages provide model Systems for the analysis of the mechanisms governing the expression of cell surface antigens specific for a particular differentiation pathway 13, 4, 121. In this study, the expression of antigens encoded by the human major histocompatibility (HLA) complex and of molecules present on myeloid cell types were analyzed with monoclonal antibodies on two somatic cell hybrids, HP1 and PUTKO 1. Somatic cell hybrid analyses of hematopoietic differentiation.

A differentiated cell expresses an entire set of specialized features. Somatic cell hybridization provides a method to examine control of gene regulation. We studied the expression of tissue-specific features in hybrids between human promyelocytes (HL-60) and human Burkitt's lymphoma cells (P3HR-1). Two hybrid lines, HP-1 and HP-2, and 18 hybrid clones were established and confirmed by karyotyp...

Differentiation of a human myeloid cell line (HL-60) toward granulocyte- and macrophage-like cells: comparison of cell surface antigen expression.

Human hematopoietic pathways have been defined mainly by the analysis of cellular morphological changes because of the paucity of other meaningful markers. Monocolonal antibodies against human cell surface antigens offer the possibility of following changes in the expression of these molecules, in particular when combined with the use of cell lines able to differentiate in vitro if provided wit...

PROGNOSTIC VALUE OF MYELOID ANTIGEN EXPRESSION IN ACUTE LYMPHOBLASTIC LEUKEMIA

Expression of cell surface molecules associated with lymphoid and myeloid lineage differentiation on the blasts of 53 patients with acute lymphoblastic leukemia (ALL) was investigated. 1.9% of cases were only HLA"7DR+ 7.6% were HLA-DR+, CDI9+ 22.6% were HLA-DR+, CDI9+, CDI0+ 30.2% were HLADR+, CDI9+, CDlO±, CD20+ and 37.7% were HLA-DR±, CD7+, CD5±. Aberrant expression of one or more myeloi...

Expression of human myeloid-associated surface antigens in human-mouse myeloid cell hybrids.

Hybrid cell lines were obtained after fusion of mouse myeloid cells (WEHI-TG) with leukocytes from two patients with chronic myeloid leukemia. A third fusion was carried out with leukocytes from a patient with acute lymphocytic leukemia. All three patients carried the Philadelphia chromosome (Ph1) in the leukemia cell population. Cytochemical analysis confirmed the myelo-monocytic nature of the...

Allele Frequencies of HLA-A, B and DRB1 among People of Fars Ethnicity Living in Tehran

Background: Human leukocyte antigens (HLA) are polymorphic cell surface proteins. Distribution of HLA alleles vary among different racial and ethnic populations in unrelated stem cell registries. Determination of HLA allele frequencies in different ethnic groups is useful for population genetic analyses. Materials and Methods: Based on data available from the Iranian Stem Cell Donor Registry...
